Abstract

Hairy root cultures represent a versatile system applicable to a wide range of studies, including the functional characterization of genes and their regulatory elements. In our laboratory, we have established a CRISPR-based genome editing protocol for oilseed rape (Brassica napus) using hairy roots. This transformation system offers a straightforward and rapid approach to evaluate the functionality and efficiency of diverse CRISPR editing constructs. Using an optimized regeneration protocol, we are able to produce transgene-free, genome edited plant lines. Our future objectives involve the application of CRISPR technologies for base editing, prime editing, and transcriptional regulation within the Brassicaceae family using hairy root cultures.
